PHARMACEUTICAL DEVELOPMENT OF EXTRACTS BASED ON CRATAEGUS MONOGYNA FRUITS AND EXPERIMENTAL SUBSTANTIATION OF THEIR REDOX-MODULATING PROPERTIES

Free-radical oxidation is regarded as one of the universal mechanisms of cell damage involved in the pathogenesis
of cardiovascular, oncological and metabolic diseases, which makes the search for natural redox modulators a practically
relevant task of pharmaceutical science. Hawthorn (Crataegus monogyna Jacq.) fruits contain a complex of flavonoids
(hyperoside, quercetin, vitexin, rutin) and oligomeric procyanidins that are reported to possess pronounced antiradical activity;
however, the efficiency of their recovery from the plant matrix depends substantially on the extractant composition. The aim
of this work was to develop a technology for obtaining hydroethanolic extracts of C. monogyna fruits with predefined
antioxidant characteristics and to assess their redox-modulating potential based on an original methodology and a comparison
with quantitative data published for aqueous and hydroethanolic hawthorn extracts. Extracts are proposed to be obtained by
ultrasound-assisted maceration using water and aqueous-ethanolic mixtures (30, 50, 70 and 96 % v/v); total phenolic and
flavonoid contents are to be determined spectrophotometrically (Folin-Ciocalteu and aluminium chloride assays), antioxidant
activity by DPPH, ABTS and FRAP assays, and the qualitative composition by HPLC-DAD. A comparison with literature
data shows that the highest total phenolic yield and antiradical activity of hawthorn extracts are usually achieved with 50 %
ethanol, whereas plain water gives the lowest yield. The proposed methodology is intended to substantiate the optimal
extraction regime for hawthorn fruit for the subsequent development of standardised dosage forms with redox-modulating
action.
